Opuntia ficus-indica extract (often abbreviated OFI or OFIE) is derived from a cactus and has been studied for its effects on inflammatory skin conditions, particularly psoriasis. Here’s what the research shows:
#1: Barrier restoration and psoriasis: When formulated into a topical cream alongside xyloglucan and pea proteins (XPO), the extract significantly reduced *S. aureus* adhesion and skin invasion in lab-grown skin cells, and restored the structural integrity of the outermost skin layer (Int J Mol Sci, 2023). In mice with psoriasis-like skin inflammation, XPO reduced redness, inflammatory markers, and abnormal skin thickening—performing better than the steroid dexamethasone in that model (Int J Mol Sci, 2023).
In a human clinical trial of 42 patients with mild-to-moderate plaque psoriasis over 28 days, the XPO formulation achieved similar improvements to a prescription combination (calcipotriol and betamethasone) in reducing disease severity, redness, itching, and scaling, with an “excellent tolerability profile” reported (J Clin Aesthet Dermatol, 2024). #2: Antioxidant mechanism: The extract activates a cellular defense pathway called AHR-NRF2-NQO1 signaling (J Med Food, 2015). In plain terms: when keratinocytes (the main cell type in your outer skin layer) were exposed to inflammatory triggers or oxidative stress, OFIE extract activated protective proteins that reduced damaging free radicals. Importantly, this effect was blocked when the key protein NRF2 was removed from cells, confirming this is the operative mechanism (J Med Food, 2015).
The same signaling also upregulated two important barrier proteins—filaggrin and loricrin—which are often depleted in barrier-disrupted skin disorders (J Med Food, 2015). #3: Wound repair acceleration: In lab studies using chitosan-based wound dressings loaded with Opuntia extract, skin cells (keratinocytes) migrated approximately 3-fold faster across a scratch wound when the extract was present compared to unloaded films, suggesting accelerated repair activity (Carbohydr Polym, 2021). Limitation: The human psoriasis trial involved only 42 patients over 28 days, so larger and longer studies would strengthen confidence in these findings. Most supportive data comes from the extract combined with other ingredients (xyloglucan and pea protein) rather than opuntia alone.
