🔬 The Science

Acetyl cysteine is a modified amino acid—one of the building blocks of protein. When your body processes NAC, it converts it into glutathione, which is an endogenous antioxidant (meaning your body naturally produces it). This matters because antioxidants neutralize free radicals, which are unstable molecules that damage skin cells and contribute to aging. #1: Antioxidant activity. In a human skin study published in the Journal of Investigative Dermatology, researchers found that NAC could be converted into glutathione and helped reduce harmful reactive oxygen species caused by ultraviolet (UV) radiation in living human skin. #2: Protection against UV-induced photoaging (sun damage and wrinkles).

The same study showed that NAC inhibited one of the key signaling pathways (extracellular signal-regulated kinase) that UV light activates to break down collagen in skin—the protein that keeps skin firm. Importantly, NAC also helped prevent UV-induced production of collagenase, an enzyme that degrades collagen. However, NAC did not prevent all UV damage pathways; it blocked some but not others (it inhibited extracellular signal-regulated kinase but not c-jun N-terminal protein kinase activation). Critically, NAC did not act as a sunscreen and had no effect on UV-induced skin redness, so it cannot replace sun protection (Journal of Investigative Dermatology, 2003). #3: Wound healing support.

In a diabetic rat model, both topical (applied to skin) and systemic (taken orally) NAC improved wound healing. Wounds treated with NAC showed smaller wounded areas and better epithelialization (skin regrowth) by day 14 compared to untreated wounds. This effect appeared linked to NAC’s ability to reduce oxidative stress in both tissue and blood (Wounds, 2019). #4: Potential skin protection against certain toxins. In a mouse model of chemically-induced skin tumors, topical NAC (applied alone or combined with vitamin E) provided significant protection against papillomatous growth (abnormal skin growths). This involved reducing free radical damage and restoring antioxidant enzyme activity in skin (Nutrition and Cancer, 2013).

#5: Antibacterial and anti-inflammatory properties in dental contexts. A 2022 systematic review in BMC Oral Health analyzed 15 studies on NAC’s use in endodontics (root canal treatment). In laboratory studies, NAC showed antibacterial and anti-biofilm efficacy comparable to or better than chlorhexidine, sodium hypochlorite, and calcium hydroxide. One clinical trial found NAC’s anti-inflammatory effect superior to calcium hydroxide. However, the review emphasized that these are mostly in vitro (lab) results and robust human clinical trials are still needed before NAC can be routinely used in clinical dental practice. Important limitations: Most evidence comes from laboratory studies and animal models. Human skin studies are extremely limited—only one published human skin study directly tested NAC on living skin (for UV protection).
