What we know about (20S)-Protopanaxadiol, or PPD, comes from a single laboratory study published in Chemistry & Biodiversity (2020) examining how this compound affects human gastric cancer cells grown in dishes. #1: PPD reduced the growth of these cancer cells more effectively than a related compound called Rg3 (Chem Biodivers, 2020). Researchers used multiple techniques to measure cell viability and observe cellular changes under microscopes. #2: The compound appeared to work by damaging two critical cellular structures. First, PPD caused problems with mitochondriaβthe cell’s energy-producing powerhousesβby generating excessive reactive oxygen species (ROS, which are chemically reactive molecules) and disrupting normal energy metabolism (Chem Biodivers, 2020).
Second, PPD damaged lysosomes, which are cellular compartments containing digestive enzymes; the researchers observed increased pH (making them less acidic) and membrane rupture in treated cells (Chem Biodivers, 2020). #3: PPD also blocked autophagy flux, a cellular recycling process, leading to accumulation of cellular debris (Chem Biodivers, 2020).
