Coffea arabica extract (CAE) is a polyphenol-rich ingredient derived from coffee plants. The research on this ingredient focuses on three main skin concerns, though it’s important to note that most studies were conducted in laboratory settings or on mice rather than human skin. #1: Antioxidant activity. CAE contains compounds like chlorogenic acid and caffeic acid that work to neutralize harmful free radicals—unstable molecules that damage skin cells. In lab tests (Int J Mol Sci, 2017), CAE demonstrated a 94.7% reducing power and 20.3% hydroxyl radical scavenging activity at specific concentrations, meaning it successfully neutralized these damaging molecules. This antioxidant capacity appears to be the foundation for its other benefits.

#2: Protection against UV damage. When skin is exposed to ultraviolet B (UVB) light, it triggers a cascade of inflammatory responses and breaks down collagen through enzymes called matrix metalloproteinases (MMPs). Research in mice (Int J Mol Sci, 2017) showed that CAE reduced UVB-induced reactive oxygen species generation in skin cells, inhibited inflammation markers like interleukin-6, and reduced MMP-1 expression—all of which contribute to photoaging. A separate study (Food Chem Toxicol, 2011) confirmed that CAE inhibited MMP-1, MMP-3, and MMP-9 expression while stimulating type I procollagen (collagen building blocks), suggesting it may help maintain skin structure when exposed to UV.

#3: Support for compromised skin barriers. In models of atopic dermatitis—a condition involving barrier dysfunction and inflammation (Int J Mol Sci, 2023)—CAE reduced transepidermal water loss (TEWL, a measure of how much moisture escapes through the skin) and restored levels of filaggrin and claudin-1, which are proteins critical to skin barrier function. CAE also reduced redness and pro-inflammatory factors in affected skin. #4: Anti-inflammatory effects. Across all three studies, CAE consistently inhibited the NF-κB pathway, a master control switch for inflammatory responses in cells. This suppression reduced the production of pro-inflammatory molecules like interleukin-1β and interleukin-6. The mechanism appears to work by blocking oxidative stress, which feeds inflammatory signaling.

Limitations: The vast majority of this research was performed in cell cultures or animal models (primarily mice), not on human skin. Only one study (Int J Mol Sci, 2017) specifically mentions human fibroblasts in vitro, but even this is laboratory-based rather than a clinical trial on living human skin. Human studies would be needed to confirm these benefits translate to real-world skincare results.
