🔬 The Science

Glutathione is a tripeptide—a small protein made of three amino acids—that exists inside your cells in a reduced form, where it helps maintain what’s called ‘redox balance,’ essentially protecting cells from oxidative stress (Indian J Dermatol Venereol Leprol, 2016). #1: Skin-lightening mechanism. Glutathione may reduce skin darkening by inhibiting tyrosinase, an enzyme involved in melanin production, and by shifting the type of melanin your skin makes from eumelanin (darker) to phaeomelanin (lighter) (Indian J Dermatol Venereol Leprol, 2016). However, #2: the evidence for how well this works in real people remains mixed. Three randomized controlled trials support that topical and oral glutathione have skin-lightening effects with a good safety profile, but the 2016 review notes key questions remain unanswered about how long treatment should last, whether effects persist after stopping, and what maintenance looks like (Indian J Dermatol Venereol Leprol, 2016).

#3: Topical application. Among topical forms, 0.5% glutathione was significantly more effective than 0.1% or placebo (Int J Dermatol, 2025). A 2% topical formulation also showed ability to brighten skin color in sun-exposed areas when measured by melanin index (J Cosmet Dermatol, 2019). Notably, combining topical 2% glutathione with oral glutathione produced better results than either alone (Int J Dermatol, 2025). #4: Oral supplementation. Five randomized controlled trials found that oral glutathione at doses of 250 mg once daily, 250 mg twice daily, or 500 mg once daily significantly reduced the melanin index compared to placebo (Int J Dermatol, 2025).

A 12-week study using 250 mg/day of either the reduced or oxidized form of oral glutathione showed a tendency toward lower melanin index and UV spots, with some sites showing significant wrinkle reduction (Clin Cosmet Investig Dermatol, 2017). However, absorption and bioavailability of oral glutathione remain limited concerns (J Cosmet Dermatol, 2019). #5: Location matters. Skin-lightening effects appear primarily in sun-exposed areas; no significant differences were observed in sun-protected areas (J Cosmet Dermatol, 2019). #6: Intravenous form not recommended. Despite popularity in some regions, there is no evidence that intravenous glutathione is effective for skin lightening, and adverse effects led the Philippine FDA to issue a public warning against its off-label use for this purpose (Indian J Dermatol Venereol Leprol, 2016).

Only one placebo-controlled study on IV glutathione exists, showing weak results, and IV glutathione is now considered contraindicated (Int J Dermatol, 2025). #7: Safety profile. Topical and oral glutathione show minimal to no serious adverse effects in the studies reviewed (Indian J Dermatol Venereol Leprol, 2016; Clin Cosmet Investig Dermatol, 2017; Int J Dermatol, 2025). Some mild nonserious adverse events have been reported but are uncommon (J Cosmet Dermatol, 2019). #8: Sustainability questions. Both topical and oral glutathione provide what researchers describe as ‘moderately efficacious’ skin-lightening but are ‘unsustainable,’ with variable costs and unclear long-term maintenance needs (Int J Dermatol, 2025).

The 2016 review calls for larger, longer-term studies with better-defined outcomes before firm conclusions can be drawn (Indian J Dermatol Venereol Leprol, 2016), and a 2019 systematic review concludes the overall evidence remains ‘inconclusive’ (J Cosmet Dermatol, 2019).
