Glycosaminoglycans are long-chain sugar molecules that play important roles in skin structure and hydration. Proteoglycans are proteins with glycosaminoglycans attached to them—think of them as core proteins decorated with these sugar chains. Cells need inorganic sulfate (a form of sulfur) to build these molecules properly. Research in the Journal of Biochemical and Biophysical Methods (1988) examined what happens when cells try to synthesize these molecules without sulfate present. The study compared mouse cartilage cells working under sulfate-free conditions versus normal physiological sulfate levels. The findings showed two key issues: #1: Cells produced significantly less glycosaminoglycan when sulfate was absent (J Biochem Biophys Methods, 1988), and #2: The glycosaminoglycans that were made had lower sulfation—meaning fewer sulfur-containing groups attached—and these lower-sulfated versions were not produced at all under normal sulfate conditions (J Biochem Biophys Methods, 1988).
The researchers concluded that sulfate-free conditions should be avoided in laboratory studies because they don’t reflect how these molecules are actually made in the body. This is important context for understanding sulfate-free skincare claims, though the available research focuses on laboratory conditions rather than consumer skincare applications.
