1-Naphthaleneacetic acid (NAA) is a synthetic compound that mimics auxin, a natural plant hormone. While plants naturally use auxin and its receptors for growth regulation, mammalian cells—including human cells—don’t have these natural auxin receptors, yet research suggests NAA can still influence them in unexpected ways. In human cell culture research (Adv Biol, 2024), #1: NAA demonstrated up to a 2-fold increase in the yield of cultured human cells across multiple cell types tested, including both established cell lines and primary cells. Importantly, #2: this boost in cell numbers did not result from faster cell division—flow cytometry analysis showed NAA did not alter the cell cycle.
Instead, #3: the mechanism appears to involve rescuing cells from death, as apoptosis-specific staining indicated reduced cell death in NAA-treated cultures. Gene sequencing revealed that #4: a gene called ANGPTL4, which is involved in lipid metabolism and natural cell survival pathways, was activated by NAA treatment, and this gene was required for NAA’s beneficial effects. In agricultural applications, NAA has been used for decades to regulate plant growth. Research on strawberries (J Agric Food Chem, 2019) showed that #5: NAA regulates growth through multiple interconnected metabolic pathways, and the timing of application matters—spraying NAA at different ripening stages produced opposite effects on citric acid levels and related gene expression.
A separate study on plant root development (J Agric Food Chem, 2020) found that chemically modified versions of NAA—caged compounds designed to release slowly under ultraviolet light—could #6: stimulate secondary root growth at concentrations where free NAA was actually toxic to plants, and these modified compounds showed no significant toxicity to animal cell cultures at those same concentrations. Current research remains limited to laboratory and agricultural settings; human skin applications have not been studied in these sources.
