Hexapeptide-11 is a six-amino-acid peptide with the structure Phe-Val-Ala-Pro-Phe-Pro (FVAPFP), originally discovered in yeast extracts and now produced through solid-state chemical synthesis (Redox Biol, 2015). In laboratory testing on human fibroblasts—the cells that make up much of your skin’s structural tissue—Hexapeptide-11 triggered several protective cellular mechanisms. #1: It activated the proteasome and autophagy, which are your cells’ cleanup systems that remove damaged proteins and debris (Redox Biol, 2015). #2: It increased production of chaperone proteins, which help other proteins fold correctly and function properly (Redox Biol, 2015). #3: It boosted antioxidant response genes, helping cells defend against free radical damage (Redox Biol, 2015).

#4: Specifically, it caused increased nuclear accumulation of Nrf2, a key regulator of antioxidant defenses (Redox Biol, 2015). When fibroblasts were exposed to oxidative stress—a major driver of premature cellular aging—Hexapeptide-11 provided significant protection against senescence, the process where cells stop dividing and functioning well (Redox Biol, 2015). In an actual human skin study, the peptide improved skin elasticity, a measure of skin’s ability to stretch and bounce back (Redox Biol, 2015). The research also noted that Hexapeptide-11 activated matrix metalloproteinases (MMPs), enzymes involved in skin remodeling, while simultaneously suppressing cell migration (Redox Biol, 2015).
