Menadione is a member of the class of 1,4-naphthoquinones that is 1,4-naphthoquinone which is substituted at position 2 by a methyl group. It is used as a nutritional supplement and for the treatment of hypoprothrombinemia. It has a role as an EC 3.4.22.69 (SARS coronavirus main proteinase) inhibitor, an antineoplastic agent, an angiogenesis inhibitor, a nutraceutical and a human urinary metabolite. It is a member of 1,4-naphthoquinones and a vitamin K. A synthetic naphthoquinone without the isoprenoid side chain and biological activity, but can be converted to active vitamin K2, menaquinone, after alkylation in vivo. menadione has been reported in Streptomyces, Thermoplasma acidophilum, and other organisms with data available.

In research published in Antibiotics (Basel) in 2024, behenic acid demonstrated the ability to inhibit biofilm formation by Staphylococcus aureus—a bacterium that naturally lives on human skin—at concentrations up to 20 mg/L. Importantly, it achieved this inhibition without actually killing the bacteria cells themselves, which is potentially valuable because it allows for physical removal of problematic bacteria while reducing the selective pressure that drives antibiotic resistance.

The mechanism appears related to behenic acid’s surfactant properties (similar to the common emulsifier Tween 80), rather than disruption of typical bacterial cell processes like hydrophobicity or polysaccharide production. #2: Formulation stability in oleogels. Two separate studies in the Journal of Colloid Interface Science (2020) and Journal of Cosmetic Science (2021) examined behenic acid as a key component in oleogel systems—oil-based gels created by mixing behenic acid with behenyl alcohol in various vegetable oils. These formulations are used in cosmetic products for hair and skin applications. The research found that specific weight ratios between behenic acid and behenyl alcohol create more stable, harder gels with better oil-binding capacity, though the optimal ratio varies slightly depending on which oil base is used (sunflower, apricot, and rapeseed oils favor a 7:3 ratio, while olive and camelina oils favor 8:2).

This research is technical and addresses formulation engineering rather than direct skin benefits. Limitations: The biofilm research is limited to laboratory studies on one bacterial species; human skin studies are not present in these sources.
